چکیده

The isoprenoids, cholesterol and rubber, are known to arise from mevalonic (3,5-dihydroxy-3-methylvaleric) acid (5, 7). The same is true of carotenes (1, 6). Thus it has been found in this laboratory that mlevalonic acid is rapidly incorporated into carotenes and related compounds in the tonmato fruit (6). The hydrocarbon fraction of tomatoes previously incubated with 2-C'4-mevalonate contains radioactivity, however, not only in the known carotenes but also in a previously undescribed, colorless polyene which possesses absorption maxima at 208 my and 231 my. The high specific activity of this compound, which has been referred to as fraction II (6), suggests that it may be a precursor of carotenes. This paper reports further experiments pertaining to the identity of fraction IT and to its role in the biosylntlhesis of carotenoids.

Retinol and ?-Carotene inhibit PTZ-induced kindling in mice

Vitamin A and its derivatives have recently reported to be implicated in synaptic plasticity. The possible effect of vitamin A and its precursor, ?-carotene, on seizure acquisition was tested in PTZ kindling model of epilepsy. Vitamin A and ?-carotene were tested for their ability to 1) suppress seizures (clonic and tonic) and lethality induced by PTZ in PTZ-kindled mice (anticonvulsant effect)...

On the biosynthetic relationship between lycopene and colorless polyenes in tomatoes.

The suggestion of Zechmeister (1) that phytofluene might be a precursor of lycopene has been expanded by Porter and Lincoln (2), who proposed on the basis of experiments with tomatoes that a successive dehydrogenation of more saturated polyenes’ leads to the biosynthesis of lycopene and other carotenoids. This sequence would involve the intermediary formation of tetrahydrophytoene, phytoene, ph...

Terpenoid biosynthesis from 1-deoxy-D-xylulose in higher plants by intramolecular skeletal rearrangement.

The incorporation of [1-13C]- and [2,3,4,5-13C4]1-deoxy-D-xylulose into beta-carotene, lutein, phytol, and sitosterol in a cell culture of Catharanthus roseus was analyzed by NMR spectroscopy. The labeling patterns of the isoprene precursors, isopentenyl pyrophosphate and dimethylallyl pyrophosphate, were obtained from the terpenes by a retrobiosynthetic approach. 13C Enrichment and 13C13C coup...

Structural studies on bacterial carotenoids and their biosynthetic implications.

Detailed studies of the properties of carotenoids isolated from diphenylamine-inhibited cultures of Rhodospirillum rubrum have revealed a number of novel structures which indicate new features of carotenoid biosynthesis in the photosynthetic bacteria. The dehydrogenation of phytoene to coloured carotenoids occurs by a sequence which is different from that in higher plants in that 7,8,11,12-tetr...
